1. Etiology of brittle diabetes: The total lack of insulinogenic reserve is most likely to be the foundamental etiologic factor in diabetic brittleness. Inappropriate glucagons secretion may also be responsible for diabetic instability.
2. Is gut GLI one of incretin candidates? Gut GLI exists in the intestinal mucosa and is secreted into the circulation, which hold true for the characteristics of incretin. But, no insulinogenic activity is found in gut GLI. Based on these findings, it can be concluded that GLI is not incretin.
3. Occasionally, iodinated glucagons treated with chloramine-T does not react to an antibody specific for glucagons, which appears due to oxidation of the methionine residue at position 27 in the molecule. Inclusion of dimethyl sulfoxide in the chloramine-T reaction results in preservation of the immunoreactivity of the treated preparations.
